Saracens' Owen Farrell will make his first start at fly-half for England against Wales after Charlie Hodgson was ruled out with a finger injury.
The 20-year-old will get his chance at ten due to his team-mate's injury on Saturday at Twickenham.
Brad Barritt switches to his favoured inside centre position while David Strettle continues on the wing.
Mouritz Botha keeps his place in the second row and Matt Stevens is again available from the bench.
Interim coach Stuart Lancaster said: "Charlie cut his finger in training earlier in the week and needed stitches but it swelled up and there is a sprain there.
"We assessed him this morning and he cannot grip fully. Owen has trained throughout and knows our patterns and plays so we are fully confident in his ability to run the game from ten and we are looking forward to seeing how Brad and Manu [Tuilagi] gel together in the centre."
